Poly (3-hydroxybutyrate) (P(3HB)) is the most studied thermoplastic biopolymer belonging to the polyhydroxyalkanoate (PHA) family, the main features of which include rapid biodegradability and biocompatibility. The bioplastic recovery process is an important step during production and can directly influence the characteristics of PHAs. However, more efficient methods for the production of P(3HB) are necessary to make it economically viable. The aim of the present study was to improve the standard, chloroform-based, extraction step for the recovery of P(3HB). The polymer was produced using a Ralstonia solanacearum strain. The following parameters were improved in the recovery process: heating time, separation method (filtration or liquid-liquid phase separation), biomass state (fresh or dry cell concentrate) and the solvent:biomass ratio. By improving the chemical extraction of P(3HB) we recovered 98% of the cumulative polymer and reduced the heating time by 75%. Furthermore, we improved the separation process and developed an extraction solution that was faster and more economical.

Six plant growth promoting rhizobacteria isolates - two Streptomyces (DFs1296 and DFs1315), three Bacillus (DFs1414, DFs1420 and DFs1423) and one Pseudomonas (DFs1421) - were used to microbiolize tomato seeds for the control of bacterial spot caused by Xanthomonas gardneri. Three assays were conducted in a completely randomized design with six replications. In each assay, X. gardneri suspensions (10(8)CFUmL-1) were spray-inoculated on the leaves. In the first and second assays, three and four leaves, respectively, were assessed for disease severity. In the third assay, three leaves per plant were assessed for the number of lesions per leaf, leaf area and dry weight. Results showed that one of the Bacillus sp. isolate (DFs1420) consistently provided the greatest relative reduction (48%) of tomato bacterial spot severity.

A busca por alternativas ao uso intensivo de agrotóxicos no controle de doenças tem recebido grande atenção da pesquisa agrícola. Rizobactérias têm reconhecida capacidade de reduzir doenças em diversas culturas e promover de crescimento de plantas. O objetivo deste trabalho foi avaliar a eficácia de seis isolados de rizobactérias, pré-selecionadas, no controle de Ralstonia solanacearum e Fusarium oxysporum f. sp. lycopersici (FOL), em casa de vegetação e relacionar este comportamento a produção de compostos "in vitro". Foi avaliada a capacidade de estas rizobactérias produzirem quitinases, amilases, lipases, compostos antibióticos e de solubilizar fosfato. A microbiolização das sementes com a rizobactéria DFs1421 (Pseudomonas sp.) reduziu os valores de AACPD de murcha bacteriana em ambos os ensaios (36,6 e 91,7% no primeiro e segundo ensaios respectivamente). Este controle pode ser associado à produção de compostos responsáveis pela antibiose observada "in vitro". Isolados de Streptomyces (DFs1296 e DFs1315), bem como de Bacillus (DFs1414) e o indutor químico ASM reduziram significativamente a murcha de fusário, variando de 22,5 a 76%. O controle observado por parte das rizobactérias pode ser atribuído à atividade quitinolítica e/ou antibiótica por compostos voláteis.

Replacing the intensive use of pesticides for alternative methods in disease control has been an important aim for agricultural research. Rhizobacteria are known to be capable of reducing diseases levels in many crops and also of promoting plant growth. The objective of this work was to evaluate the efficacy of six pre-selected rhizobacteria isolates in controlling Ralstonia solanacearum and Fusarium oxysporum f. sp. lycopersici (FOL) under controlled conditions in a greenhouse, and to link such capacity to the "in vitro" production of selected biologically active compounds. The ability of these rhizobacteria isolates to produce chitinases, amylases, lipases, antibiotic compounds was investigated. Additionally, their ability in solubilizing phosphate was also checked. Microbiolization of seeds with one rhizobacterium DFs1421 (Pseudomonas sp.) reduced the tomato wilt AUDPC in both assays (36.6 and 91.7% in the first and second assays respectively). Such efficacy in wilt control is likely to be linked with the production of antibiotics as observed "in vitro". Streptomyces (DFs1296 and DFs1315) and Bacillus (DFs1414), and the chemical inducer (ASM) reduced significantly fusarium wilt ranging 22.5 to 76%. This may be owing to the observed chitinolytic activity and / or antibiosis in the presence of volatile compounds.

Avaliou-se o potencial de oito isolados de rizobactérias que eficientemente controlam doenças fúngicas em arroz [DFs185 (Pseudomonas synxantha), DFs223 (P. fluorescens), DFs306 (não identificado), DFs416, DFs418 e DFs419 (Bacillus sp.), DFs422 (Bacillus subtilis), DFs471 (Stenotrophomonas malthophilia)] no controle de Meloidogyne graminicola, in vitro, e em arroz irrigado em casa de vegetação. As rizobactérias DFs185, DFs223, DFs306 e DFs416 se destacaram na redução da eclosão e no aumento da mortalidade de J2 de M. graminicola. Todas as rizobactérias foram capazes de produzir pelo menos um composto associado ao biocontrole de nematoides (quitinases, fosfatases, lipases, proteinases e sideróforos). O número de galhas e de ovos de M. graminicola em plantas de arroz foi reduzido em relação às plantas vindas de sementes não microbiolizadas. O fator de reprodução do nematoide foi reduzido, em média, em 29%, com destaque para as rizobactérias DFs185 e DFs223 que proporcionaram redução de 35%. Esse resultado é significativo, porque são rizobactérias que também controlam doenças fúngicas.

The potential of eight rhizobacteria that effectively control fungal diseases in rice [DFs185 (Pseudomonas synxantha), DFs223 (P. fluorescens), DFs306 (not identified), DFs416, DFs418 e DFs419 (Bacillus sp.), DFs422 (Bacillus subtilis), DFs471 (Stenotrophomonas malthophilia)] was evaluated for the control of Meloidogyne graminicola in vitro and in irrigated rice in the greenhouse. The rhizobacteria DFs185, DFs223, DFs306, DFs416 and DFs419 exceeded in reducing hatching and in increasing the mortality of M. graminicola J2. All rhizobacteria were able to produce at least one compound associated with the biocontrol of nematodes (chitinasis, phosphatases, lipases, proteinasis and siderophores). The number of eggs and galls of M. graminicola in rice plants was reduced compared to plants from seeds not microbiolized. The nematode reproduction factor was reduced on average by 29%, highlighting rhizobacteria DFs185 and DFs223 that reduced 35%. This result is significant because the rhizobacteria also control fungal diseases.

O potencial dos isolados DFs185 (Pseudomonas synxatha), DFs223 (Pseudomonas fluorescens), DFs306 (não identificado), DFs416, DFs418 e DFs419 (Bacillus sp.), DFs422 (Bacillus subtilis) e DFs471 (Stenotrophomonas maltophilia) foi avaliado para o controle da mancha-parda (Bipolaris oryzae) e da escaldadura (Gerlachia oryzae) do arroz. Para tanto, sementes da cultivar El Passo L144 foram imersas em suspensão (10(7) a 10(8) UFC/mL) de cada um dos isolados, agitadas por 30 min a 10ºC e semeadas em vasos, sendo utilizado o delineamento inteiramente casualizado com quatro repetições. Foram utilizadas duas testemunhas: uma tratada com solução salina e outra com solução salina mais fungicida (Carboxin+Thiran). Realizaram-se dois ensaios para cada um dos patógenos, inoculados nas plantas por aspersão, e avaliados quanto à severidade aos 7, 14 e 21 dias após a inoculação calculando a área abaixo da curva de progresso da doença. Para mancha-parda, no primeiro ensaio, o melhor desempenho foi proporcionado pelos isolados DFs185, DFs223 e DFs306 que apresentaram 80, 86 e 70% de redução da severidade. No segundo ensaio, o isolado DFs306 foi o mais eficiente deles, com redução na severidade da doença chegando a 74% em relação à testemunha, sendo que os isolados DFs223, DFs185 e DFs306 proporcionaram incremento na massa de grãos em 74, 70 e 59%, respectivamente. Para o controle da escaldadura, no primeiro ensaio, os isolados que apresentaram melhores resultados foram DFs416 e DFs418 com 65 e 59% de controle, respectivamente. No segundo ensaio, se destacaram os isolados DFs416 e DFs185 reduzindo a severidade da doença em 64 e 60%, respectivamente sem incrementar a produção.

The potential of isolates DFs185 (Pseudomonas synxatha), DFs223 (Pseudomonas fluorescens), DFs306 (unidentified), DFs416, DFs418 and DFs419 (Bacillus sp.), DFs422 (Bacillus subtilis) and DFs471 (Stenotrophomonas maltophilia) was evaluated for the control of rice brown spot (Bipolaris oryzae) and leaf scald (Gerlachia oryzae). Rice seeds cv. El Passo L144 were immersed in a suspension (10(7) a 10(8) CFU/mL) of each of the isolates, agitated for 30 min at 10ºC and sowed in pots, in complete randomized design with four replications. Two controls were used: one with seeds treated with saline solution, and the other with saline solution plus fungicide (Carboxin+Thiran). Two assays for each of the pathogens were carried out. Plants were inoculated by spraying and their disease severity was evaluated at 7, 14 and 21 days after inoculation for the calculation of the area under disease progress curve. The best result for brown spot biocontrol in the first assay was provided by isolates DFs185, DFs223 and DFs306, which presented 80, 86 and 70% of control. In the second assay, isolate DFs306 was the most efficient, with reduction in disease severity reaching 74% in relation to control. The isolates DFs223, DFs185 and DFs306 provided a rise in grain yield of 74, 70 and 59%, respectively. For the control of leaf scald, in the first assay, the isolates that presented the best results were DFs416 and DFs418, which showed 65 and 59% of control, respectively. In the second experiment, isolates DFs416 and DFs185 reduced the disease severity, respectively, by 64 and 60%, without increasing the yield.

Coletaram-se, de diferentes cidades do estado do Rio Grande do Sul (RS), 62 e 194 amostras de sementes e grãos de arroz da safra 2000-2001 e safra 2001-2002, respectivamente. Amostras das localidades de Agro Reis, Agudo, Alegrete, Arambaré, Arroio Grande, Bagé, Caçapava do Sul, Cachoeira do Sul, Cachoeirinha, Camaquã, Canguçu, Capão do Leão, Capivari do Sul, Dom Pedrito, Eldorado do Sul, Esteio, Guaíba, Herval, Itaqui, Júlio Castilhos, Morro Redondo, Palmares do Sul, Pedro Osório, Pelotas, Pinheiro Machado, Porto Alegre, Quaraí, Restinga Seca, Rio Grande, Rosário do Sul, Santa Vitória do Palmar, Santa Rita, Santo Ângelo, Santo Antônio, São Borja, São Gabriel, São Lourenço do Sul, São Luiz Gonzaga, Tapes, Turuçu, Uruguaiana e Viamão foram avaliadas quanto à ocorrência da praga quarentenária ausente no país, Helicoceras spp.. Foram examinadas 400 sementes por amostra, pelo método do papel filtro. O fungo Helicoceras oryzae foi encontrado em três amostras da safra 2001/2002: 268 (EEA-406), 647 (FORMOSA) e 652 (IRGA 419), de Cachoeirinha. Este levantamento permitiu constatar a presença de H. oryzae., no Brasil. Considerado patógeno de pouca importância, o Comitê de Sanidade Vegetal do Conesul estuda excluí-lo da lista de pragas quarentenárias. Estudos futuros para avaliação da disseminação e impactos da doença são necessários para subsidiar medidas de controle efetivas.

Samples of rice seeds and grains were collected from different localities in the state of Rio Grande do Sul during 2000-2001 (62 samples) and 2001-2001 (194 samples). Samples from the localities of Agro Reis, Agudo, Alegrete, Arambaré, Arroio Grande, Bagé, Caçapava do Sul, Cachoeira do Sul, Cachoeirinha, Camaquã, Canguçu, Capão do Leão, Capivari do Sul, Dom Pedrito, Eldorado do Sul, Esteio, Guaíba, Herval, Itaquí, Júlio Castilhos, Morro Redondo, Palmares do Sul, Pedro Osório, Pelotas, Pinheiro Machado, Porto Alegre, Quaraí, Restinga Seca, Rio Grande, Rosário do Sul, Santa Vitória do Palmar, Santa Rita, Santo Ângelo, Santo Antônio, São Borja, São Gabriel, São Lourenço do Sul, São Luiz Gonzaga, Tapes, Turuçu, Uruguaiana and Viamão were evaluated for the presence of a pest not found in Brazil, Helicoceras spp. Using the blotter test, 400 seeds were examined for each sample. The fungus H. oryzae was found in three samples of the 2000-2001 season: 268 (EEA-406), 647 (FORMOSA) and 652 (IRGA 419) from Cachoeirinha. These results allowed confirmation of the presence of the fungus H. oryzae. The Conesul Committee of Plant Health and Safety is evaluating the exclusion of this fungus, considered a pest of minor importance, from the quarantine list. Future studies about dissemination and impacts of the disease are necessary to support effective control measures.

Objetivou-se avaliar o efeito de oito isolados bacterianos pré-selecionados de Pseudomonas synxatha, P. fluorescens, Bacillus subtilis, Bacillus sp. e Stenotrophomonas malthophilia no controle da queima-das-bainhas do arroz, causada por Rhizoctonia solani. Sementes da cultivar El Passo L144 foram imersas em suspensão (A540=0,5) de cada um dos isolados e agitadas por 30 min a 10ºC. Sementes imersas somente em solução salina e em salina mais fungicida (Carboxin + Thiran) foram utilizadas como testemunhas. Foram semeadas 10 sementes por vaso, em quatro repetições, dispostas em delineamento completamente casualizado. Foram realizados três ensaios, sendo que no primeiro foi possível selecionar três isolados como promissores, com reduções na severidade da doença atingindo 50, 33,3 e 16,7 %, respectivamente. Estes isolados foram utilizados nos ensaios posteriores, instalados em casa de vegetação e conduzidos até o ponto de colheita, onde foi possível observar o efeito biocontrolador propiciado, principalmente, pelo isolado de P. fluorescens DFs223, com reduções significativas na severidade da doença chegando a 88 e 91,7% no segundo e terceiro ensaios respectivamente. Em ambos os ensaios, houve incremento tanto do número de panículas quanto do número de perfilhos e da massa seca de raízes em até 42,8, 81,2 e 113% respectivamente, nas plantas tratadas com o isolado de P. fluorescens DFs223.

Eight pre-selected bacterial strains were evaluated for control of rice sheath blight caused by Rhizoctonia solani, Pseudomonas synxatha, P. fluorescens, Bacillus subtilis, Bacillus sp. and Stenotrophomonas malthopilia. Rice seeds cv. El Passo L144 were treated by immersion in a bacterial suspension (A540=0.5) of each isolate under agitation at 10ºC for 30 min. Seeds immersed in saline solution and in saline plus fungicide (Carboxin + Thiran) were used as control. Ten seeds per pot were used, in four replications, in a completely randomized design. Three assays were performed, and in the first it was possible to select the three strains which reduced the severity of the disease, reaching 50, 33.3 and 16.7%, respectively. These isolates were used in the two last assays, which were maintained in greenhouse until the harvesting stage, where it was possible to observe the biocontrol effect, mainly by the strain of P. fluorescens DFs223, with significant reduction in the disease severity, reaching 88 and 91.7% in the second and third assays, respectively. There was an increase in the number of panicles, as well as in the number of tillers and dry mass of roots, reaching to 42.8, 81.2 and 113 % respectively, in plants treated with this isolate in both assays.

Tem-se dado muita ênfase ao controle biológico mediante seleção de microorganismos. Porém, para se avaliar o potencial de biocontroladores de forma massal e em pequeno intervalo de tempo é necessário desenvolver um bioensaio eficiente. Bactérias de diferentes sítios, num total de 760 isolados, foram selecionadas para compatibilidade com Rhizobium leguminosarum bv. phaseoli estirpes SEMIA 4077 e SEMIA 4080, onde 596 isolados foram inefetivos contra ambos rizóbios. Sementes de feijão foram imersas em suspensão de cada um destes isolados sendo agitadas por 5 horas a 10ºC, plantadas em solo não esterelizado, sendo as plantas mantidas em casa de vegetação. Após o desenvolvimento das folhas cotiledonares e folhas primárias, estas foram retiradas e avaliadas por bioensaio para o controle de Xanthomonas axonopodis pv. phaseoli (XAP). Nas folhas cotiledonares, observou-se que o isolado DFs093, proporcionou 100% de controle, DFs041 e DFs1297 propiciaram, 90% e DFs490, DFs769, DFs831, DFs842 e DFs843 proporcionaram 80% de controle. Nas folhas primárias, o isolado, DFs482 propiciou 100% e os isolados DFs080, DFs348, DFs513, DFs622, DFs769, DFs842 e DFs912 proporcionaram 80% de controle para XAP.

Emphasis has been given on selection of micro-organism for biological control. However, in order to evaluate the biological control potential of a great number of micro-organisms in a small period of time it is necessary to develop an efficient bioassay. Seven hundred and sixty bacterial isolates from different habitats, were selected for compatibility with Rhizobium leguminosarum bv. phaseoli (SEMIA 4077 e SEMIA 4080). Among them 596 isolates were ineffective against both rhizobia. Bean seeds immersed in suspension of each one of these isolates were agitated for 5 hours at 10ºC and sowed in non-sterilized soil. The plants were kept in greenhouse. After the development of cotyledonary and primary leaves, these were removed and bioassayed for Xanthomonas axonopodis pv. phaseoli (XAP) control. In the cotyledonary leaves, it was observed that the isolate DFs093 offered 100% control, DFs041 and DFs1297 offered 90% and DFs490, DFs769, DFs831, DFs842 and DFs843 offered 80% control. In the primary leaves, the DFs482 isolated offered 100% and the DFs080, DFs348, DFs513, DFs622, DFs769, DFs842 and DFs912 offered 80% of XAP control.
